Unsubscribe rate is one of the clearest signals that something about a specific email campaign - content, frequency, or targeting - is misaligned with what your list actually wants to receive.
Worked example
For 20 unsubscribes out of 10,000 emails delivered:
Unsubscribe Rate = 20 / 10000 x 100 = 0.2% (Acceptable but worth monitoring)

A sudden spike above your normal baseline for one specific campaign usually points to something particular about that send (unexpected content, too-frequent messaging, or a poorly matched segment) rather than a broader ongoing list quality problem.
