Total DNA Yield (ng) = Concentration (ng/┬ÁL) x Elution Volume (┬ÁL)

Example

50 ng/┬ÁL concentration, eluted in 100 ┬ÁL:

Total Yield = 50 x 100 = 5,000 ng (5.0 ┬Ág)

Why This Conversion Matters

Downstream molecular biology applications - PCR, sequencing library preparation, restriction enzyme digests - typically specify how much total DNA mass is required per reaction, not concentration alone. Knowing your total yield tells you exactly how many downstream reactions your extracted sample can support, which is essential for planning experiments and avoiding running out of precious sample mid-protocol.

A Practical Note

Spectrophotometer (UV absorbance) readings can be affected by contaminants like residual protein or phenol from the extraction process, sometimes inflating apparent concentration - fluorometric quantification methods that bind specifically to double-stranded DNA are generally considered more accurate for precise downstream applications where exact DNA quantity matters most.
